MPG® Glyceryl has a non-ionic hydrophilic coating providing two adjacent hydroxyl groups for the covalent attachment of biomolecules containing primary and/or secondary amines. It is used for coupling antigens or antibodies for specific antibody or antigen capture when low non-specific binding is desirable. Biomolecules are covalently coupled to the MPG® Glyceryl in a two-step procedure which begins with periodate oxidation of the hydroxyl groups on the particle surface to form aldehydes. The amine-containing biomolecule is then covalently linked to the aldehyde-modified MPG® by reductive amination. Since the surface hydroxyl groups are non-ionic, non-specific binding to the particle surface is minimized.
Following the suggested protocol, coupling efficiencies of up to 75 µg of antibody per mg of MPG® Glyceryl are achieved.
