Isolate pure, intact yeast or gram positive bacterial genomic DNA 10 to 20 times faster than other standard protocols with our new DNA-Pure Yeast Genomic Kit. This high yielding system is based on a unique lysis buffer technology. There is no need for lytic enzymes and lengthy laborious procedures such as enzyme digestions and organic extractions, which may result in variable quality DNA that is often sheared. The protocol is a rapid, 30 minutes, and results in the isolation of high quality, intact yeast genomic DNA up tp 200 Kb.
This system works well for lysing thick cell walls, allowing you to grow your yeast cells to early stationary phase to maximize DNA yield. This kit has been tested successfully with a variety of yeast strains including Saccharomyces cerevisiae, Schizosaccharomyces pombe and Pichia pastoris. In addition, the kit has been tested on strains with very tough cell walls such as Candida famata. Typical yields from this kit are 10 - 20 µg from a 1.5 - 3 ml culture. The DNA obtained using the DNA-Pure Yeast Genomic Kit gives excellent restriction digestion (see figure) and PCR results.

This kit also works to successfully isolate gram+ bacterial genomic DNA from Bacillius subtilis, Bacillus anthracis and staphylococcus aureus. Typical yields of approximately 10µg of pure bacterial DNA are obtained from 1.5ml overnight cultures.
The DNA-Pure Yeast Genomic Kit utilizes an easy to follow protocol which consists of the following steps: pellet cells; resuspend cells and add lysis solution; add protein precip solution after a 10 minute incubation at 70°C; centrifuge and then isopropanol precipitate DNA from the supernatant; wash pellet and resuspend in TE or water. The kit comes complete, there are no hidden costs.
