Researcher: I am using your DNA-Pure™ Yeast Genomic Kit to purify yeast DNA. After performing the cell lysis step my solution still appears cloudy. My cells don't appear to be lysing completely. Should I increase lysis time?

TechTALKer:Unlike some other protocols, the purification technique used in this kit does not rely on complete breakdown of the yeast cell wall and nuclear membrane. Thus, the solution will not clear and will remain cloudy after the lysis step. This is expected. However, if you know the yeast type you are using has particularly tough cell walls, then you can increase the lysis step by 5 or 10 minutes.
